    Protection by 20-5,14-HEDGE Against Surgically-Induced Ischemia Reperfusion Lung Injury in Rats

    Background We previously reported that the cytochrome P450 product 20-hydroxyeicosatetraenoic acid has prosurvival effects in pulmonary artery endothelial cells and ex vivo pulmonary arteries. We tested the potential of a 20-hydroxyeicosatetraenoic acid analog N-[20-hydroxyeicosa-5(Z),14(Z)-dienoyl]glycine (20-5,14-HEDGE) to protect against lung ischemic reperfusion injury in rats. Furthermore, we examined activation of innate immune system components, high mobility group box 1 (HMGB1) and toll-like receptor 4 (TLR4), in this model as well as the effect of 20-5,14-HEDGE on this signaling pathway. Methods Sprague-Dawley rats treated with 20-5,14-HEDGE or vehicle were subjected to surgically induced, unilateral lung ischemia for 60 minutes followed by reperfusion for 2 hours in vivo. Injury was assessed histologically by hematoxylin and eosin, and with identification of myeloperoxidase immunohistochemically. The HMGB1 and TLR4 proteins were identified by Western blot. Caspase 3 activity or 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, a yellow tetrazole, incorporation were used to measure apoptosis and cell survival. Results The ischemia reperfusion injury evoked atelectasis and hemorrhage, an influx of polymorphonuclear cells, and increased TLR4 and HMGB1 expression. Caspase 3 activity was increased, and 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide incorporation was decreased. The 20-5,14-HEDGE protected against each of these endpoints, including infiltration of polymorphonuclear cells, with no changes in caspase 3 activity in other organs. Conclusions Lung ischemia reperfusion produces apoptosis and activation of the innate immune system including HMGB1 and TLR4 within 2 hours of reperfusion. Treatment with 20-5,14-HEDGE decreases activation of this response system, and salvages lung tissue

Thus, the use of artificial intelligence in agriculture can be a promising solution to ensure sufficient food supply on a global scale. To investigate the state-of-the-art in the use of artificial intelligence-based systems in agriculture, we provide a structured literature review. We show that research has been done in the field of irrigation and plant growth. In this regard, camera systems often provide images as training/input data for artificial intelligence-based systems. Finally, we provide a research agenda to pave the way for further research on the use of artificial intelligence in sustainable agriculture

    The Effects of Wearable Health Technology on Cancer Survivors’ Physical Activity, Sedentary Behavior, and Weight: A Meta-Analysis

    Wearable health technology (WHT) has been suggested as a health intervention in preventing or reducing health risk factors in clinical populations. Cancer survivors exhibit risk factors prior to cancer diagnosis and acquire comorbidities as a result of their treatment. These conditions may increase likelihood of cancer recurrence and reduce quality of life by inhibiting survivors’ physiological function and predisposing them to other maladies. Several studies have suggested WHTs as an intervention in mitigating these risks. However, there lacks a comprehensive review of the current evidence to determine the efficacy of WHT interventions. Thus, a literature search of WHT studies within cancer survivors was conducted and a meta-analysis was performed on the four most commonly reported health outcomes, namely weight, step count, moderate-to-vigorous physical activity (MVPA) and sedentary time. A meta-analysis was performed to determine if positive or negative effects between WHT and these outcomes exist and what the effect sizes were. Twelve papers met inclusion criteria. Of the ten studies that measured MVPA, a moderate positive effect was found between WHT interventions and weekly MVPA. Five studies measured average daily step count and from this, a weak positive effect was found between WHT interventions and step count increase. Five studies measured sedentary time and revealed a weakly positive correlation between WHT use and increased sedentary time, though this data was unreliable. Lastly, three studies measured weight and revealed a weak negative effect between WHT use and weight, however, this was also limited due to the small sample. Thus, this review validates the use of WHT to improve MVPA and possibly step count in cancer survivors while it appears WHTs have a lesser impact on weight and potentially an opposite effect on sedentary time

    An expanded multilocus sequence typing scheme for propionibacterium acnes : investigation of 'pathogenic', 'commensal' and antibiotic resistant strains

    The Gram-positive bacterium Propionibacterium acnes is a member of the normal human skin microbiota and is associated with various infections and clinical conditions. There is tentative evidence to suggest that certain lineages may be associated with disease and others with health. We recently described a multilocus sequence typing scheme (MLST) for P. acnes based on seven housekeeping genes (http://pubmlst.org/pacnes). We now describe an expanded eight gene version based on six housekeeping genes and two ‘putative virulence’ genes (eMLST) that provides improved high resolution typing (91eSTs from 285 isolates), and generates phylogenies congruent with those based on whole genome analysis. When compared with the nine gene MLST scheme developed at the University of Bath, UK, and utilised by researchers at Aarhus University, Denmark, the eMLST method offers greater resolution. Using the scheme, we examined 208 isolates from disparate clinical sources, and 77 isolates from healthy skin. Acne was predominately associated with type IA1 clonal complexes CC1, CC3 and CC4; with eST1 and eST3 lineages being highly represented. In contrast, type IA2 strains were recovered at a rate similar to type IB and II organisms. Ophthalmic infections were predominately associated with type IA1 and IA2 strains, while type IB and II were more frequently recovered from soft tissue and retrieved medical devices. Strains with rRNA mutations conferring resistance to antibiotics used in acne treatment were dominated by eST3, with some evidence for intercontinental spread. In contrast, despite its high association with acne, only a small number of resistant CC1 eSTs were identified. A number of eSTs were only recovered from healthy skin, particularly eSTs representing CC72 (type II) and CC77 (type III). Collectively our data lends support to the view that pathogenic versus truly commensal lineages of P. acnes may exist. This is likely to have important therapeutic and diagnostic implications

    Biomarkers for Radiation Pneumonitis Using Noninvasive Molecular Imaging

    Our goal is to develop minimally invasive biomarkers for predicting radiation-induced lung injury before symptoms develop. Currently, there are no biomarkers that can predict radiation pneumonitis. Radiation damage to the whole lung is a serious risk in nuclear accidents or in radiologic terrorism. Our previous studies have shown that a single dose of 15 Gy of x-rays to the thorax causes severe pneumonitis in rats by 6–8 wk. We have also developed a mitigator for radiation pneumonitis and fibrosis that can be started as late as 5 wk after radiation. Methods: We used 2 functional SPECT probes in vivo in irradiated rat lungs. Regional pulmonary perfusion was measured by injection of 99mTc-macroaggregated albumin. Perfused volume was determined by comparing the volume of distribution of 99mTc-macroaggregated albumin to the anatomic lung volume obtained by small-animal CT. A second probe, 99mTc-labeled Duramycin, which binds to apoptotic cells, was used to measure pulmonary cell death in the same rat model. Results: The perfused volume of lung was decreased by about 25% at 1, 2, and 3 wk after receipt of 15 Gy, and 99mTc-Duramycin uptake was more than doubled at 2 and 3 wk. There was no change in body weight, breathing rate, or lung histology between irradiated and nonirradiated rats at these times. Pulmonary vascular resistance and vascular permeability measured in isolated perfused lungs ex vivo increased at 2 wk after 15 Gy of irradiation. Conclusion: Our results suggest that SPECT biomarkers have the potential to predict radiation injury to the lungs before substantial functional or histologic damage is observed. Early prediction of radiation pneumonitis in time to initiate mitigation will benefit those exposed to radiation in the context of therapy, accidents, or terrorism

    Crack propagation through phase separated glasses: effect of the characteristic size of disorder

    We perform fracture experiments on nanoscale phase separated glasses and measure crack surface roughness by atomic force microscopy. The ability of tuning the phase domain size by thermal treatment allows us to test thoroughly the predictions of crack font depinning models about the scaling properties of crack surface roughness. It appears that in the range of validity of these depinning models developed for the fracture of brittle materials, our experimental results show a quantitative agreement with theoretical predictions: beyond the characteristic size of disorder, the roughness of crack surfaces obeys the logarithmic scaling early predicted by Ramanathan, Ertas and Fisher (PRL97
